[目的]研究健脾化瘀解毒方对胃癌前病变（PLGC）大鼠胃黏膜组织上皮细胞的影响。[方法]将SD大鼠分为6组：正常组、模型组、维酶素组及健脾化瘀解毒方低、中、高剂量组。采取1-甲基-3-硝基-1-亚硝基胍（MNNG）溶液灌胃构建PLGC大鼠模型，健脾化瘀解毒方低、中、高剂量组每天按3.75 g/kg、7.5 g/kg、15 g/kg的剂量灌胃给药，维酶素组每天按0.2 g/kg的剂量灌胃给药，正常组和模型组灌胃给予等容量生理盐水。使用光学显微镜观察胃黏膜组织病理学，使用透射电镜（TEM）分析胃黏膜组织上皮细胞的超微结构。[结果]正常组大鼠胃黏膜正常。模型组大鼠全部表现为慢性萎缩性胃炎病理状态，伴肠上皮化生与不典型增生。维酶素组和健脾化瘀解毒方低、中、高剂量组大部分表现为慢性萎缩性胃炎病理状态，但肠上皮化生及不典型增生情况较模型组减少。在胃黏膜组织上皮细胞超微结构方面，正常组细胞核为圆形或椭圆形，细胞器形态正常，细胞质密度高；模型组细胞核为不规则形，大且深染，核质比增大，细胞器显著减少，胞质出现裂隙，细胞间联系不发达；维酶素组细胞核呈类椭圆形，细胞器减少，胞质出现裂隙；健脾化瘀解毒方各剂量组细胞核呈类椭圆形，细胞器形态正常，细胞质密度高。[结论]健脾化瘀解毒方可改善PLGC大鼠胃黏膜组织病理损伤，改善胃黏膜组织上皮细胞的超微结构，提示健脾化瘀解毒方对PLGC有较好的治疗效果。

[Objective]To investigate the effects of Jianpi Huayu Jiedu Decoction on gastric mucosal epithelial cells in rats with precancerous lesions of gastric cancer(PLGC).[Methods]SD rats were randomly divided into 6 groups: normal group, model group, Vitacoenzyme group, and low-, medium-, and high-dose Jianpi Huayu Jiedu Decoction groups. The PLGC rat model was established by intragastric administration of 1-methyl-3-nitro-1-nitrosoguanidine(MNNG)solution. Rats in the low-, medium-, and high-dose Jianpi Huayu Jiedu Decoction groups were given intragastric administration of the decoction at doses of 3.75 g/kg, 7.5 g/kg, and 15 g/kg per day, respectively. Rats in the Vitacoenzyme group were intragastrically administered with vitacoenzyme at a dose of 0.2 g/kg per day. Rats in the normal group and model group were given an equal volume of normal saline by gavage. Pathological changes of gastric mucosa were observed under optical microscope, and the ultrastructure of gastric mucosal epithelial cells was analyzed by transmission electron microscopy(TEM).[Results]The gastric mucosa of rats in the normal group was normal. All rats in the model group presented with chronic atrophic gastritis combining intestinal metaplasia and dysplasia. Most of the rats in the Vitacoenzyme group and low-, medium-, and high-dose Jianpi Huayu Jiedu Decoction groups suffered from chronic atrophic gastritis, but the incidences of intestinal metaplasia and dysplasia were lower than those in the model group. In terms of the ultrastructure of gastric mucosal epithelial cells, the nuclei in the normal group were round or oval, with normal organelle morphology and high cytoplasmic density. In the model group, the nuclei were irregular,large and deeply stained, with an increased nuclear-cytoplasmic ratio, significantly reduced organelles, cytoplasmic clefts, and underdeveloped intercellular connections. In the Vitacoenzyme group, the nuclei were sub-oval, with reduced organelles and cytoplasmic clefts. In each dose group of Jianpi Huayu Jiedu Decoction, the nuclei were sub-oval,with normal organelle morphology and high cytoplasmic density.[Conclusion]Jianpi Huayu Jiedu Decoction can alleviate the pathological damage of gastric mucosa and improve the ultrastructure of gastric mucosal epithelial cells in PLGC rats, suggesting that it has a good therapeutic effect on PLGC.
